چکیده

Berberine, the plant alkaloid, was recently shown to induce self-structure in poly(A) by a unique dilution technique. The interaction of berberine and five 9-(u-amino) alkyl ether analogues with polyadenylic acid [poly (A)] has been studied to understand the role of an additional anchoring module in the formation and/or amplification of self-structure in poly(A). The binding was characterized by absorption and fluorescence titration, Job’s plot and isothermal titration calorimetry. All the molecules bound cooperatively to poly(A). Self-structure formation was confirmed by circular dichroic melting, optical melting, and dilution experiments. Energetics of the interaction revealed that as the alkyl chain length increased, the binding was more entropy dominated. The results showed that berberine and all the analogues induced self-structure formation in poly(A). The length of the alkyl chain had a significant influence on the ease of formation of self-structure. New insights in terms of structural and thermodynamic aspects into self-structure formation in poly(A) by berberine analogues are revealed from these studies.

Biophysical Characterization of the Strong Stabilization of the RNA Triplex poly(U)•poly(A)*poly(U) by 9-O-(ω-amino) Alkyl Ether Berberine Analogs

BACKGROUND Binding of two 9-O-(ω-amino) alkyl ether berberine analogs BC1 and BC2 to the RNA triplex poly(U)(•)poly(A)(*)poly(U) was studied by various biophysical techniques. METHODOLOGY/PRINCIPAL FINDINGS Berberine analogs bind to the RNA triplex non-cooperatively. The affinity of binding was remarkably high by about 5 and 15 times, respectively, for BC1 and BC2 compared to berberine. The s...

Study of new NBS-LRR genes analogues in cucurbits native types in Iran

Nucleotide binding site leucine-rich repeats (NBS-LRR) accounting for the main disease resistance proteins play an important role in plant defense against pathogen attack. The current study aimed to identify new NBS-LRR gene members in native types of cucurbit species in Iran. Accordingly, DNAs of melon, cucumber and cantaloupe native types to Iran were identified using three primer pairs. PCR ...

Berberine induces G1 arrest and apoptosis in human glioblastoma T98G cells through mitochondrial/caspases pathway.

Berberine is an isoquinoline plant alkaloid with a long history of being used for the treatment of many diseases in Chinese herbal medicine. Berberine has a wide range of biochemical and pharmacological effects, including antitumor activities, but its mechanism of action is not clearly understood. In this study, we investigated that the relationship between the antiproliferative activities of b...

Characterization of vacuolar transport of the endogenous alkaloid berberine in Coptis japonica.

Alkaloids comprise one of the largest groups of plant secondary metabolites. Many of them exhibit strong biological activities, and, in most cases, they are accumulated in the central vacuole of alkaloid-producing plants after synthesis. However, the mechanisms involved in alkaloid transport across the tonoplast are only poorly understood. In this study, we analyzed the vacuolar transport mecha...
